这句话是正确的.“that is on the second floor"是个定语从句.这句话还可以表达为 My office,which is on the second floor,is very small.

My office that is on the second floor is very small.这句语法没有错误。两个句子都是定语从句，用that的话，前面就不需要加逗号；用which的话，前面需要加逗号，同理，who前面也要加逗号，如That boy, who won the game, is my friend. “defing relative clause”是什么意思，多少年没碰语法，不懂了。
